The images show a 50:50 mixture of live and dead Jurkat cells that were stained with trypan blue (TB) dye. TB stains cells that have a leaky membrane (dead) whereas cells with intact cell membranes remain unstained (live). In the “Intensity Image,” dead cells appear dark (filled arrowhead) and live cells appear light (open arrowhead). The “Absorbance Image” and “Moles Image” were generated from the brightfield “Intensity Image” to determine the moles of TB inside each cell as a quantitative, traceable measure of their viability. All three images are the same field of view.
